I play e4 and my opponent plays nf6. I have never encountered this position as my brother was the first to play it. What should I do?

I suggest you look up the Alekhine’s Defense, there are a lot of lessons about this line. You should probably attack the Knight with one of these attacks, especially if your brother is not too experienced with this line:
2. e5 Nd5 3. d4 d6 4. c4 Nb6
or
2. e5 Nd5 3. c4 Nb6 4. d4 (or 4. a4)
You could also play 2. Nc3 instead of 2. e5 if you want to avoid this line entirely!
